Profile Analysis of Whole Protein oF Salmonella Typhi that Resistant and Sensitive to Chloramphenicol
ABSTRACT
Salmonella typhi consist of many species,from the antigenic scheme it is more than 200 species. Chloramphenicol as an antibiotic used for diseases treatment caused by Salmonella typhi has the ability to compete with messenger RNA in the ribosomal binding site. Chloramphenicol is reversibly attached to the 50 S ribosome at amino acid binding site where the aminoacyl-tRNA binds amino acids in the binding region, so the peptidyl transferase activy in the protein synthesis process is inhibited. The purpose of this study was to determine the profile of whole protein of Salmonella typhi that resistant and sensitive to chloramphenicol. The research approach used was post-test only design. SDS PAGE analysiswas performed to determine the whole protein profile differences of Salmonella typhi that resistant and sensitive to shloramphenicol. This research was conducted at the Biomolecular Laboratory of Veterinary Medicine Faculty (FKH), Microbiology Laboratory of Science and Technology Faculty (FSAINTEK) and Institute of Tropicaln Diseases (ITD), Universitas Airlangga Surabaya from September to October 2009.Result shows that the shorter of distance and Rf of sample, the greater of molecular weight of the protein,so that the protein profiles depending on the distance and the Rf of sample at band of electrophoresis result of 12,5% agarose gel. The conclusion of study there are defferences of whole protein profiles of Salmonella typhi that resistant and sensitive to chloramphenicol.
